L'edificio fu edificato probabilmente all'inizio del IX secolo, sotto il dogado di Pietro Tradonico o di Orso I Partecipazio (una tradizione ne fissa la fondazione all'837).
A differenza di altre chiese veneziane, San Polo non è stata mai completamente ricostruita, sicché ha conservato l'impianto originale a tre navate tipico dell'architettura veneto-bizantina. I numerosi rimaneggiamenti, invece, ne fanno un edificio poco unitario dal punto di vista stilistico.
Subì una prima ristrutturazione tra il Tre e il Quattrocento e a questo periodo risalgono gli elementi gotici, in particolare il portale laterale archiacuto, con modanature a tortiglione e ornamenti floreali, le ogive della navata laterale e il rosone.
A partire dal 1804 sino alla riconsacrazione del 1838 la chiesa ha subito i pesanti interventi progettati da David Rossi: in quell'occasione furono sostituite le colonne della navata centrale, chiuse alcune aperture per aprirne altre e venne usato, nel complesso, un anomalo stile neoclassico. L'abside fu ricostruita a pianta semiottagonale con finestre centinate. I restauri conclusi di recente hanno in parte recuperato gli elementi quattrocenteschi, in particolare il soffitto a carena di nave, e rimossi gli inutili ornamenti ottocenteschi. La chiesa ospita l'intera collezione di dipinti di TIEPOLO raffiguranti la via Crucis.

The building was probably built at the beginning of the ninth century, under the dogado of Pietro Tradonico or of Bear I Partecipazio (a tradition establishes its foundation at 837).
Unlike other Venetian churches, San Polo has never been completely rebuilt, so it has preserved the original three-aisle structure typical of the Venetian-Byzantine architecture. The many changes, however, make it a little unified building from the stylistic point of view.
It underwent a first renovation between the fifteenth and fifteenth centuries and to this period date back the Gothic elements, in particular the side portal archiacuto, with twisted moldings and floral ornaments, the ogives of the side aisle and the rose window.
From 1804 until the re-consecration of 1838, the church underwent the heavy interventions designed by David Rossi: on that occasion the columns of the central nave were replaced, some openings were opened to open others and an anomalous neoclassical style was used as a whole . The apse was rebuilt with a semi-octagonal plan with arched windows. The recently completed restorations partly recovered the fifteenth-century elements, in particular the ship's hull ceiling, and removed the unnecessary nineteenth-century ornaments.

Es erfuhr zwischen dem 15. und 15. Jahrhundert eine erste Umstrukturierung und die gotischen Elemente stammen aus dieser Zeit, insbesondere das seitliche archiacuto-Portal mit verdrehten Formteilen und floralen Ornamenten, die Ogiven des Seitenschiffs und das Rosettenfenster.
Von 1804 bis zum Wiederaufbau im Jahr 1838 wurden in der Kirche umfangreiche Eingriffe von David Rossi durchgeführt: Bei dieser Gelegenheit wurden die Säulen des Mittelschiffs ersetzt, einige Öffnungen wurden geschlossen, andere wurden geöffnet, und es wurde ein anomaler neoklassizistischer Stil als Ganzes verwendet. . Die Apsis wurde mit einem halb-achteckigen Grundriss mit Bogenfenstern umgebaut. Bei den kürzlich abgeschlossenen Restaurierungen wurden die Elemente des 15. Jahrhunderts, insbesondere die Schiffsrumpfdecke, teilweise wiederhergestellt und die nutzlosen Verzierungen aus dem 19. Jahrhundert entfernt. Die Kirche beherbergt die gesamte Sammlung von TIEPOLO-Gemälden, die die Via Crucis darstellen.
