 Fondata nel IX secolo, è una delle chiese più antiche di Venezia.Risalirebbe per tradizione al 555: in base ad un'annotazione di Marin Sanudo, che riporta la scritta 555 che appariva sulla facciata (da leggersi però 976, tenendo conto del more veneto): data assai più probabile della precedente, visto che il Lanfranchi indica che la zona fu bonificata tra il 959 e il 976. Durante i restauri del 1903 si scoprirono i resti di un edificio circolare al di fuori della chiesa (probabilmente un battistero analogo a quello di Torcello) e la data 555.
Si trovarono inoltre frammenti di pavimento, a 60 centimetri al di sotto dell'attuale superficie calpestabile, assai simili a quelli delle basiliche di San Marco e di Santa Maria Assunta di Torcello.
Tuttavia nessuna notizia archivistica è anteriore al 1000. Sanudo dice inoltre che la chiesa di san Giacomo fu fabbricata dalle famiglie Campoli di Oderzo e Da Mula dalle Contrade.

Piuttosto incerta l'etimologia: secondo alcuni derivante da una famiglia Orio, secondo altri più probabilmente da luprio, palude (e infatti l'isola era circondata da una zona paludosa poi bonificata) secondo altri ancora da un riferimento all'antistante rio, verso cui è rivolta la facciata (dal rio). Secondo il Lorenzetti l'origine del nome sarebbe legata al fatto che l'isola in cui venne edificata la chiesa era infestata dai lupi, da cui de lupio o de lupao poi storpiata in dall'Orio, oppure sia un riferimento a un albero di alloro (lauro) probabilmente situato vicino alla chiesa stessa.
Venne riedificata nel 1225 a cura delle nobili famiglie Badoer e Da Mula e rimaneggiata nel periodo gotico, tra il XV e il XVI secolo.

 Founded in the 9th century, it is one of the oldest churches in Venice. During the restorations of 1903, the remains of a circular building outside the church were discovered (probably a baptistery similar to That of Torcello) and the date 555.
There were also found fragments of floor, 60 cm below the current walkable surface, very similar to those of the Basilicas of St. Mark and of Santa Maria Assunta of Torcello.
However, no archival news is before 1000. Sanudo also says that the church of San Giacomo was manufactured by the Campoli family of Oderzo and Da Mula from the Contrada.

Rather uncertain than etymology: according to some derived from an Orio family, according to others probably luprio, marsh (and in fact the island was surrounded by a swampy area then boned), according to others still referring to the ancient rio, towards which The façade is turned (from the Rio). According to Lorenzetti, the origin of the name would be related to the fact that the island where the church was built was infested by wolves, from which lupine or lupo then crippled in the ori, or a reference to a bay tree (Lauro) probably located near the church itself.
It was rebuilt in 1225 by the noble families Badoer and Da Mula and restructured in the Gothic period between the 15th and the 16th century.
