Dieses Gebäude hat einst Leopold I. für seinen dritten Sohn Dietrich von Anhalt-Dessau bauen lassen. Nach ihm wurde es schließlich auch benannt. Der Bau dieses Gebäudes wurde nach einer Bauzeit von 5 Jahren 1752 abgeschlossen.
Nach Dietrichs Tod ging das Palais an Franz über, der es 1777 dem Philanthropinum zu Verfügung stellte. Später zog die Tochter von Leopold I. Henriette Amalie von Anhalt-Dessau für ein Jahr ein. Nach Ihrem Tod zog hier die Amalienstiftung ein.
Dieses Gebäude ist eines der wenigen, welches den 2. Weltkrieg fast unbeschadet überstanden hat.
Heute befindet sich hier die Wissenschaftliche Bibliothek der Anhaltischen Landesbücherei.
