The railway first opened in 1880 and ran between Carlsruhe and Daylesford. Stations in between included Trentham, Tylden, Lyonville, Musk Creek, Fern Hill and Bullarto. The main produce carried on the line was potatoes and firewood. Approx 50,000 passengers using the line in 1884.

In 1887 the line was expanded with a section from Ballarat to Daylesford. Stations on this line were located at: North Creswick (Junction), Broomfield, Allendale, Kingston, Newlyn, Rocky Lead, Wombat, Leonard's Hill (later renamed Leonard), Ballan road (later renamed Sailors Falls), Woodburn and Daylesford.

The Daylesford to North Creswick railway closed in July 1953.

The Carlsruhe to Daylesford railway closed to all traffic in 1978.
